Space ending?

When you build a space ship and send a colonist into space, can you play on with the rest of them?

yes.

you always have the option to keep playing.  Even if:
1) you travel to the ship
2) you build and launch your own ship
3) everyone dies (you can wait around for a wanderer, or watch the raiders obliterate your base)

The "left behind" starting colonists in A18 may add a new mechanic to this; I have no idea

The 'Left Behind' colonists in the A18 character creation screen are not colonists you previously used. They act as a group of excess colonists, so that you can swap out randomly generated colonists if you want to try and roll a new one without getting rid of the old one. I can't really explain this very well, Barky has a good video which should explain things. Link is below.
